[高血压医学论文] 【摘要】 目的: 研究mmp2和nm23在无HBV表达的人肝癌细胞株HepG2和持续表达HBV的HepG2细胞株(HepG2.215)中的表达及差异,进一步了解HBV在肝癌侵袭转移中的作用. 方法: 通过免疫细胞化学、Western Blot检测两组细胞株HepG2和HepG2.215中mmp2和nm23的表达. 结果: 肝癌细胞株HepG2.215与HepG2细胞株相比较,其mmp2表达的水平明显升高(P<0.01),而nm23表达的水平却明显下降(P<0.01). 结论: mmp2和nm23的表达水平能反映肝癌的侵袭转移能力. mmp2在HepG2.215中的高表达和nm23在HepG2.215的低表达,可提示长时间的HBV感染可能会改变转移相关基因的表达,且有可能会导致肝癌细胞扩散转移.

【Abstract】 AIM: To investigate the expression of nm23 and mmp2 in HepG2 cells and HepG2.215 cells which persistently expresses hepatitis B virus by integrated HBV genome with HepG2, and to further understand the function of HBV in invasion and metastasis of hepatocellular carcinoma (HCC). METHODS: The expressions of mmp2 and nm23 proteins were determined by immunocytochemistry and western blot in the HCC cell lines. RESULTS: Compared with HepG2 cells, the expression of mmp2 protein increased, and that of nm23 protein decreased in HepG2.215 cells. CONCLUSION: The expressions of nm23 and mmp2 protein in HCC may reflect the metastasis potential of hepatocellular carcinoma. There is a marked difference in the expression rate of nm23 to mmp2 between the HepG2.215 cells and HepG2 cells. The results indicate a possibility that long time HBV infection could induce the expression profile of metastasis……
